Abstract: Cell Preservation Services Inc. (CPSI) is a research company that focuses on metabolic pathways triggered in human cells due to short term and long-term exposures to various stressors, including the cold. CPSI has discovered and subsequently defined the molecular actions of a number of adjuvants that when combined with freezing can increase the lethal effect of cold several fold ? a research program that is coupled to CPSI?s cryosurgery division. CPSI has also set forth a new paradigm that hypothesizes that (1) cells activate different sets of cell-specific stress pathways when exposed to the cold and, (2) some of these pathways compete with each other with some cell survival and cell death pathways being activated simultaneously. CPSI?s research approach is dedicated to understanding these pathways which once fully defined, may lead to better ablation and storage methodologies. This Phase 1 project will use a variety of airway cells and define those metabolic and molecular pathways that are activated when airway cells are chilled. The Specific Aims include tasks that will use cDNA microarrays and sh/siRNA technology to identify those pathways that are preferentially activated during cold storage in airway cells. An additional Specific Aim will be dedicated to determining the role of the mitochondria in the cell?s response to cold exposure. Finally, the JNK, AKT and bcl-2 pathways will be investigated to determine which need to be considered on the pathway to improving airway cell preservation protocols. Thus, this project is dedicated to using molecular tools so that the cell death and survival pathways activated in airway cells can be identified and ultimately manipulated to improve lung preservation/transplantation methodologies.
